WEBINAR: Titration of 164 antibodies using CITE-seq on human PBMCs
Summary:
10x Genomics and BioLegend are excited to co-host a webinar led by Felix Nettersheim, MD, postdoctoral researcher in the group of Professor Klaus Ley at La Jolla Institute for Immunology, San Diego. Dr. Nettersheim’s studies focus on understanding how metabolic reprogramming of immune cells affects atherosclerosis, the major underlying pathology of cardiovascular disease. Single cell RNA-sequencing (scRNA-seq) has emerged as a powerful tool to characterize immune cell populations. Yet, mRNA levels correlate poorly with expression of surface proteins, which are well established in defining immune cell types. CITE-seq (cellular indexing of transcriptomes and epitopes by sequencing) utilizes oligonucleotide-labeled antibodies to simultaneously analyze surface phenotypes and transcriptomes in the same cells, helping researchers overcome this issue.
Register for this webinar to learn how to titrate and choose the optimal antibody concentration for multiomics applications using CITE-seq. Dr. Nettersheim will describe studies that can be used as a resource for building an informative and cost-effective panel of TotalSeq™ antibodies, which can then be used at their optimal concentrations in future CITE-seq experiments.
Speaker: Felix Nettersheim, MD, Postdoctoral Fellow, Klaus Ley Lab, La Jolla Institute for Immunology

Multiomics and TotalSeq™ reagents
- Incorporating antibodies into your multiomics workflow requires optimization to ensure the best results. For example, titrating individual antibodies ensures that you can clearly identify the positive population without using too much of your sequencing depth for any one target.
- BioLegend oligo-conjugated antibodies enable the detection of proteins in single cells and are compatible with a variety of single-cell platforms, including the 10x Genomics Chromium Single Cell Expression Solution and Immune Profiling Solution.
- The process of titrating and balancing a large antibody panel for a single-cell multiomics experiment can be tedious, time consuming, and costly. Remove the need for antibody optimization and minimize variability between experiments by using the pre-defined BioLegend TotalSeq™ antibody cocktails.
- Each single-use tube is designed for comprehensive immune cell phenotyping and contains carefully balanced and titrated amounts of lyophilized antibodies to help you achieve optimal results and save money.
